1023 Dengue virus serotype 1 and serotype 2 and their nonstructural protein 1 may induce discrepant activation pattern of inflammation and pyroptosis in dermal fibroblasts

Dengus, a mosquito-transmitted viral infection, is estimated to cause disease burden of 400 million people worldwide per year. This study was designed elucidate the immunopathogensis initial infection stage by infecting human dermal fibroblasts WS1 cells with DENV-1 and DENV-2. The results showed several discrepancies, which included stronger innate inflammatory response cell death in DENV-2 infected cells, higher level pyroptosis cells. Reduced DENV-mediated caspase-1 IL-beta cleavage pathway both inhibitor Ac-YVAD-cmk imipramine were found, also downregulated DENV virion production Moreover, diverse activation levels death, response, at different time points found be elicited NS1 proteins. In summary, all these may suggest discrepant pattern cellular immunopathogensis, apoptosis fibroblast DENV-1, their NS1. And responses contribute replication fibroblasts.
